Analyses of T-cell receptor VB families (VB families) are most often performed to examine this diversity and the trend is to make the methodology more specific and easy to use. The aim of our study was to join this effort and to establish a method for specific and simple determination of the VB family usage. Reverse transcription PCR which allows detecting of all VB family members was combined with capillary electrophoresis in the Agilent 2100 Bioanalyzer in order to determine the relative expression values of VB families in PBMC population. Healthy donors were characterized by expression of all VB families tested, while multiple myeloma patients showed zero expression of several VB families. Described method will be used for studying of patients immune status during therapy and this further study should confirm the role of the method in monitoring T-cell population diversity in clinical settings.
