TY - CONF ID - 873202 AU - Mikalová, Lenka - Strouhal, Michal - Matějková Pospíšilová, Petra - Šmajs, David - Weinstock, George M. PY - 2009 TI - Whole genome amplification and analysis: genome differences in the genus Treponema. KW - Trepomema pallidum KW - whole genome fingerprinting KW - tpr genes N2 - Genome comparison of 6 treponemal strains including Mexico A, DAL-1 (T. pallidum subsp. pallidum), Gauthier, CDC-2 (T. pallidum subsp. pertenue), Bosnia A (T. pallidum subsp. endemicum) and Fribourg-Blanc (unclassified simian isolate) with the sequenced genome of the syphilis spirochete TPA strain Nichols. The whole genome fingerprinting technique was used for identification of heterologous regions in the chromosomal DNA of analyzed strains. All analyzed strains contained two regions of repetitive sequences (TP0433-434, TP0470) with strain specific repeat numbers. In the yaws and simian strain genomes, three specific indels were observed. Strain specific changes comprised e. g. insertion of 58 bp (TP0136) in DAL-1, deletion of 48 bp (TP0548) and insertion of 0.5 kb (TP0695-697) in Fribourg-Blanc. Genomes of T. p. pallidum, T. p. pertenue, T. p. endemicum and unclassified simian isolate are closely related and most of the observed sequence differences are localized in tpr genes and in their vinicity.
