Abstract
Treponema paraluiscuniculi is noncultivable species not infectious to humans but causing venereal spirochetosis in rabbits. The genome structure and sequence of T. paraluiscuniculi is closely related to the genome of Treponema pallidum subspecies pallidum, the syphilis causing spirochete. The genome sequence of the bacterium Treponema paraluiscuniculi strain Cuniculi A was determined by pyrosequencing, Solexa sequencing and by Comparative Genome Sequencing strategy. The assembly of the complete genome sequence was verified by whole genome fingerprinting analysis. Most conserved genes in the Cuniculi A and Nichols genomes encoded enzymes involved in general metabolic pathways and genes involved in DNA replication and maintenance and in transcription and translation processes. More than 16% of the Cuniculi A and Nichols genes encoded sequentially identical proteins, 74% genes encoded proteins with minor sequence changes and 10% of genes contained major sequence changes including frameshifts, internal stop codons and indels. The genes in the latter group encoded several Tpr proteins, recombinase proteins, components of cell envelope, antigens and regulatory and chemotactic proteins.
