Abstract
We analyzed transactivation of several p53-target genes by 23 temperature-dependent p53 mutants and stratified them into four functional groups. Seventeen mutants exhibited temperature-dependency and discriminative character in human and yeast cells. Our analysis showed that functional analysis of separated alleles in yeast is valuable tool for assessment of the human p53 status.
Abstract (in Czech)
Provedli jsme analýzu transaktivace několika cílových genů proteinu p53 celkem 23 teplotně závislými mutanty p53 a tuto sbírku jsme stratifikovali do 4 funkčních skupin. Sedmnáct mutantů vykazovalo teplotní závislost a diskriminativní povahu v jak lidských tak kvasinkových buňkách. Tato studie prokázala, že funkční analýza separovaných alel v kvsinkách je užitečným nástrojem pro studium funkčního statutu lidského proteinu p53.
