Abstract
The aim of this study was to develop a reliable and sensitive method for determination of potential biomarkers in human plasma for early GVHD detection. Blood plasma was immunodepleted by MARS Hu-14 to remove fourteen most abundant proteins. The immunodepleted plasma samples were fractionated by LC capillary system on a reverse phase (mRP C18, Agilent) in water/acetonitrile gradient and subsequently digested with trypsin. The resulting peptides were analyzed using LC-MS/MS in autoMS/MS mode.

Cílem práce bylo navrhnout a optimalizovat spolehlivou a citlivou metodu pro stanovení potencionálních biomarkerů v lidské plazmě pro časnou detekci GVHD. Plazma byla zbavena 14 nejvíce zastoupených proteinů pomocí kolonky MARS Hu-14, po té byly vzorky frakcionovány na reversní fázi na LC. Frakce byly digestovány trypsinem a analyzovány pomocí nanoLC-MS/MS.
