Abstract
A new method for determination of trace elements in submicroliter sample volumes using substrate-assisted laser desorption inductively-coupled plasma mass spectrometry (SALD ICP MS ) is presented. In contrast to conventional laser ablation (LA) ICP MS, SALD ICP MS allows quantitative analysis of metals in liquid samples because of complete sample desorption, which can be achieved using a few pulses with spot-to-spot reproducibility below 6 %. Detailed characterization, such as laser power density, raster and point desorption optimization etc. will be discussed. The performance of off-line coupling capillary electrophoresis and SALD ICP was demonstrated on speciation of 1 pg of Cr(III) and Cr (VI). SALD ICP MS can be used for the determination of trace elements in minute amounts of biological samples at physiological level as demonstrated on the determination of copper in myeloid leukemia cells to investigate the role of copper ions in cytotoxicity regulation with disulfiram .
