Abstract
Translační iniciační komplex eIF4F hraje zásadní roli v rezistenci melanomu k inhibitorům BRAF a MEK. Jeho funkce v dosud neléčených buňkách melanomu však zůstává podstatně méně objasněna. Naše výsledky ukazují, že inhibice eIF4F v těchto buňkách zvyšuje aktivitu obou sledovaných signálních drah, ERK i AMPK, přestože je dráha ERK již aktivována v důsledku onkogenní mutace. Abstract
The eIF4F translation initiation complex plays a critical role in melanoma resistance to BRAF and MEK inhibitors. Its function in treatment-naïve cells has been less well studied. We find that in these cells, eIF4F inhibition raises the activity of both signaling pathways examined in this thesis – ERK and AMPK signaling.
