Interaction of MAP2c with SH3 domains of proteins
Interakce MAP2c s SH3 doménami proteinů
Abstract
Mikrotubuly vázající protein 2c (MAP2c …more
Abstract
Microtubule associated proteins (MAPs) are intrinsically disordered proteins (IDPs) that are known to play a role in the regulation of microtubule stability and dynamics. However, they also interact with other proteins at different sites and with different affinities. Determining their structural motifs, binding partners, and regulatory pathways could help us gain a deeper understanding of their functions …more

Microtubule-associated Proteins (MAPs) regulate the stability of the microtubule. The brain cytoskeletal MAPs, MAP2 and Tau, are naturally disordered proteins, and differ by their localization, MAP2 localizes in dendrites and Tau is in axons. We study the smallest functional MAP2 isoform, MAP2c. MAP2c interacts with several brain proteins, such as 14-3-3, and plectin. In addition, MAP2c interacts with the SH3 domains of Abl and Fyn kinase with different affinities, and at different sites.
We have several constructs of SH3 domains of proteins involved in cytoskeleton organization (Vav, Nck, spectrin, cortactin). While spectrin interacts with the homolog of MAP2 in C. elegans, the interaction of MAP2c with the other proteins has not been studied yet. The goal of the thesis will be to express the different SH3 domains and test their binding to MAP2c using microscale thermophoresis. For proteins showing interaction with MAP2c, the interaction site on MAP2c will be determined using NMR.
