The focus of this diploma thesis is optimization of capillary isoelectric focusing method (CIEF) for its future utilization in the two-dimensional proteomics analysis. The thesis deals with the capillary coating proto-col and optimization of the separation of tryptic digest of several pro-teins including BSA, myoglobin, and caseins. In the last part, the possi-bility of CIEF-based sample fractionation is tested.
