Syllabus
  • The first part of laboratory training course is devoted to the work with recombinant plasmid DNA. The second part is devoted to the DNA/DNA hybridisation. The content of laboratory training course is following: Cultivation of bacterial cells carrying recombinant plasmid DNA pUC19. Small scale isolation of recombinant plasmid DNA. Electrotransformation of recombinant and nonrecombinant plasmid DNAs into bacterial cells E. coli JM 109. Selection of transformants. Chromatographic purification of recombinant plasmid DNA probe. The labelling of plasmid DNA probe by digoxigenine using nick translation. The preparation of dot blots of bacterial DNA. The carrying out of DNA/DNA hybridisation. Detection of hybrid molecules DNA using antidigoxigenin antibody.
