Course objectives
The aim of this course is to familiarize the students with basic microscopic techniques for studies on plant and animal anatomical structure, with special emphasis on techniques suitable for practical application in their future teaching practice.
Syllabus
  • Introduction. Light Microscope: Structure and Function. Köhler Illumination. Botanical Techniques: Replicas for Study by Light Microscope, Stoma Structure and Position, Subsidiary cells. Whole Mounts of Cleared Preparations, Development of Angiosperm Embryos. Squash Preparations: Cell Cycle, Mitosis, Chromosome Counting, Chromosome Doubling. Anatomical Methods: Sectioning (Hand Sections, Fixation, Infiltration and Embedding, Microtome Sections), Staining. Phase Contrast. Documentation. Zoological Techniques: Native Preparations. Vital Staining. Whole Mounts, Sections, Squashes, Dry Mounts. Fixation. Fixatives. Staining. Aqueous Media. Non-aqueous Mounting Media.
