Course Enrolment Limitations
The course is offered to students of any study field.
Course objectives
The main objective of the course is to provide the students with the ability to
- list basic techniques of optical microscopy, imaging and explain the physical principles of these techniques
- apply the correct technique for a particular sample and examine this choice in terms of the obtained image contrast,
all for the micro structured samples in biology, medicine, mineralogy, metallurgy, material sciences etc.
Learning outcomes
Student will be able:
- understand the principles of basic optical devices;
- form a scheme of a given optical apparatus and to determine the actual path of light after the pass through;
- to list and understand the basic techniques of optical contrast in microscopes;
- to list some types of modern microscopes;
Syllabus
  • 1. Spectral composition of light, light absorption, light detectors, human eye 2. Beam splitters, reflectivity and transmissivity, total reflection, beam divergence 3. Optical imaging, thin lens, optical system, main planes, focal planes 4. Construction beams, real imaging beams, entrance diaphragm, angular aperture 5. Optical microscope, coherent and noncoherent illumination of sample, field lens 6. Limiting resolution, depth of field 7. Sample properties, absorption, double refraction, scattering, reflectance 8. Method of dark field in transmission and reflection 9. Double beam interference, time and spatial coherence, visibility 10. Imaging contrast in transmission and reflection in interference microscope. 11. Linear polarization of light, polarizer, transmission of light in double refraction materials 12. Polarization microscope, molecular structure of samples and double refraction. 13. Imaging of 3D samples, contrast by scanning methods, optical tomography 14. Principles of scanning computer optical microscopy methods

Teaching methods
Lecture will be presented on-line during the standard schedule using MSTeams software. Integral part of this course is the excersize, which will be also on-line following the lecture and during which the topics from the lectures are worked through. The content of the excersize is formed by examples of the basic ray construction for the selected imaging aparatuses and/or methods.
Assessment methods
For successful completition of the course, passing the final written test is necessary. Unless stated otherwise, 80% attendance at the exercises is mandatory.
