Course objectives (in Czech)
The main goal of this practical course is to acquaint students with the principles of light microscopy. Main imaging modalities used in life sciences research will be explained and showed in practice.
Learning outcomes (in Czech)
Students will be able to explain the basic principles and uses of light microscopy in life sciences reasearch. Students will be able to explain how to prepare samples, use microscopes and analyze data.
Syllabus (in Czech)
  • practical 8 hours in two blocks (two days = 2x8 hours):
  • Day 1: Transmitted light techniques – brightfield, phase contrast, DIC, darkfield. Sample preparation for fluorescence microscopy. Image acquisition using widefield, confocal and structured illumination microscopy (Apotome, lattice SIM).
  • Day 2: Single molecule localisation microscopy – sample preparation and image acquisition. Image analysis part – image processing, deconvolution, single molecule localisation data analysis, object segmentation and quantification.
  • The practical part can be done only if the student is enrolled in the online theoretical part - S5015 Light microscopy methods in biology.

Students perform laboratory experiments according to curriculum. Study materials are available in the IS MU.
