Course objectives
The main aim of the course is to practice the knowledge gained by the students within the Bi7015 subject with an emphasis on the analytical use of the respective properties of nucleic acids.
Learning outcomes
At the end of the course the students will be able to perform • basic nucleic acids analyzes by means of spectroscopic, electrophoretic and electrochemical methods • construction of labeled oligonucleotides using DNA polymerases and labeled nucleotides • DNA cleavage by restriction endonucleases • modification of DNA using selected chemical reagents and analysis of the products
Syllabus
  • 1. Gel electrophoresis of nucleic acids, proteins and their complexes, two-dimensional electrophoresis. 2. Blotting, protein immunodetection. 3. Magnetic separation techniques, immunoprecipitation. 4. Centrifugation methods, DNA precipitation, extraction with organic solvents, gradient centrifugation, ultrafiltration. 5. Chromatographic methods, FPLC of proteins on ionex, affinity and gel-filtration columns. 7. DNA sequencing and footprinting. 7. Electrochemical methods, determination of DNA and proteins, detection of DNA damage and interactions.
