Course objectives
Seznámit studenty s principy a aplikacemi metod genového inženýrství u rostlin. Tj. s manipulacemi s nukleovými kyselinami a rostlinami za účelem využívání těchto postupů při měnění genotypu a fenotypu rostlin a rozšiřování spektra možností využívání rostlin v teoretickém výzkumu a především zemědělské, potravinářské, farmaceutické a medicínské praxi).
Syllabus
  • 1. Plant genome structure. Plant genome complexity, nuclear, mitochodndrial and plastid DNA. Plant chromosome structure. 2. Principles of DNAs manipulations. Isolation and separation of nucleic acids Hybridization of nucleic acids. Nucleic acids sequence detection. 3. Polymerase chain reaction. Principles. Special procedures - RT-PCR, RAPD, RACE-PCR, AFL. 4. Plant transgenosis. Vector types (plasmids, fags, artificial chromosomes). Agrobacterium tumefaciens, A. rhizogenes - Ti-plazsmid, RI-plasmid a T-DNA. T-DNA transfer to plant cell. Detection of T-DNA transfer. 5. Transgenic (genetically modified) plants. Herbicide resistance. Insects resistance. Virus resistance. Increased metabolic compounds production. 6. Advantages and risks joined with the use of genetically modified plants in agriculture. Decrease of environmental stress. New therapy possibilities. Decrease of genetic variability. 7. Legislation connected to genetically modified plants Distribution of growing genetically modified plants in the world and their production. Legislation in the EU. Legislation in Czechia and Slovakia. Legislation in the other parts of world.

Tj. s principy a aplikací metod molekulární biologie u rostlin (manipulacemi s nukleovými kyselinami a jejich analýzou), strukturou rostlinného genomu jaderného a mimojaderného, detekce a izolace genů u rostlin, transgenoze rostlin (způsoby, využití v teoretickém výzkumu a zemědělské, potravinářské a farmaceutické praxi), Možnosti využití v dalších oblastech biologie rostlin.
