Rychlý rozvoj technik využívajících DNA čipy umožnil v posledních době provádění genotypizace velkého množství jednobodových polymorfismů (SNPs) v rámci GWAS (genome-wide association studies). V rámci naší studie, zabývající se analýzou SNPs asociovaných s rizikem vzniku schizofrenie, jsme provedli návrh DNA čipu pro analýzu 96 SNPs pomocí metody GoldenGate na přístroji BeadExpress reader (Illumina). Při výběru byly zohledněny výsledky posledních GWAS (Genome Wide Association Studies) studií s našimi předchozími výsledky. Celkem bylo analyzováno 405 vzorků pacientů a kontrol mužského pohlaví. Průměrná call rate u vzorků byla 98%, kdy do následné statistické analýzy byly použity pouze vzorky s call rate vyšší než 90%. Po analýze výsledků pomocí Fisherova dvoustranného testu a Bonferroniho korekci bylo nalezeno šest polymorfismů (rs6595788, rs28705343, rs3737967, rs77692880, rs9960767 a rs6104567) signifikantně asociovaných s rizikem vzniku schizofrenie (p < 0.05). Zajímavým zjištěním bylo, že z celkem 12 nejsignifikantněji asociovaných SNPs z GWAS analýz byla asociace nalezena pouze u čtyř (rs6595788, rs28705343, rs3737967 a rs9960767). Tento výsekem potvrzuje, že všechny výsledky GWAS analýz je nutné nezávisle potvrzovat v menších studiích z více specifikovanými jednici a pokud možno sledovat možný vliv pohlaví na míru asociace. |

Fast development of microarray techniques in recent years has allowed us to perform genotyping of large number of single nucleotide polymorphisms (SNPs) as a part of GWAS (genome-wide association studies). In our study we have observing SNPs associated with a risk of developing schizophrenia. We have designed DNA microarray analyzing 96 SNPs using GoldenGate method on BeadExpress reader (Illumina). In the selection we have followed the results of recent GWAS (Genome Wide Association Studies) and those from our previous studies. The whole analysis comprised 405 men samples from patients and healthy controls. The avarage call rate in our samples was 98%, while in further statistic analysis we have used only those samples with call rate above 90%. After analysis using Fisher two-tailed test a Bonferroni corrections we have found six polymorphisms (rs6595788, rs28705343, rs3737967, rs77692880, rs9960767 a rs6104567) significantly associated with a risk of developing schizophrenia (p < 0.05). Interestingly, from 12 most significanly associated SNPs from GWAS analyses there have been found association only in four SNPs (rs6595788, rs28705343, rs3737967 a rs9960767). This confirms the necessity to replicate GWAS results by conducting smaller unbiased studies with better specified individuals and to consider involvment of gender on the degree of these association. |
