TY - CONF ID - 1213309 AU - Celá, Andrea - Dědová, Tereza - Mádr, Aleš - Glatz, Zdeněk PY - 2014 TI - In-capillary derivatization of amino acid followed by capillary electrophoresis with fluorescence detection KW - capillary electrophoresis, fluorescence detection, in-capillary derivatization, amino acids N2 - In-capillary derivatization reaction offers negligible consumption of reactants and samples, high level of automation and the time between a reaction and detection of product is strictly defined. As a result, errors caused by instability of the products and various times between the reaction and analysis are eliminated. Amino acids are important compounds for a living organism and their levels in biological fluids are altered as a response to a different physiological state of the organism. Quantification of amino acids is hinders by a lack of strong chromophores in their structure thus derivatization reaction has to be performed for a sensitive detection. The subject of this study is a development of in-capillary derivatization of amino acids using naphthalene-2,3-dicarboxaldehyde, sodium cyanide and mildly alkali condition ensured by tetraborate buffer. Separation of the product of above-mentioned reaction by done using micellar electrokinetic chromatography by means of capillary electrophoresis coupled with fluorescence detection. ER -
