Wallerian degeneration is a cascade of stereotype cellular and molecular events distal to injury of nerve fibers considered to be a sort of innate immune reaction or neuroinflammation. The cellular events comprise inflammatory activation of Schwann cells and invasion of various types of immune cells including macrophages that produce cytokines and chemokines. However, little is known on beneficial effect of proinflammatory cytokines upregulated by activated Schwann cells. Therefore, the goal of present study was to investigate expression of proinflammatory cytokines (IL-1b, TNFa, IL-6) and their receptors in activated Schwann cells distal to crush and compression injury of rat sciatic nerves.The sciatic nerve sections from naïve or sham-operated rats displayed very week immunostaining for investigated cytokines and their receptors. Immunofluorescence of significantly higher intensity was found in sections distal to nerve lesion as well as in sections of contralateral sciatic nerve. Immunostaining revealed that proinflammatory cytokines and their receptors are dominantly expressed by activated Schwann cells which many of them were in close contact with growing axons. Surprisingly, activated ED-1+ macrophages in distal sciatic nerve segments displayed significantly lower level of proinflammatory cytokines and their receptors.
