Schizophrenia is a severe complex mental disorder with high heritability and prevalence 0.5–1% of the population worldwide. Reliable diagnostic tests are still unavailable and the diagnosis is based mainly on interview of the person and family members. Finding biomarkers would improve the understanding of the disease and enable diagnosis in the early onset phase. Significant differences in the amino acid profiles in patients´ plasma in comparison with healthy were noted. Capillary electrophoresis coupled with laser induced fluorescence detection is a powerful tool for analysis of amino acids in biological samples. As a result, new derivatization and separation conditions for determination of all proteinogenic amino acids in human plasma were developed. Optimization and validation of the method is the subject of this contribution.

Schizofrenie je závažná mentální porucha s vysokou dědičností a prevalencí celosvětově přibližně 0.5-1 % populace. Neexistují spolehlivé diagnostické testy a diagnóza je stále založena především na rozhovoru s postiženou osobou a členy rodiny. Objev biomarkerů by pomohl objasnit toto onemocnění a umožnil jeho včasnou diagnózu. Byly zaznamenány významné rozdíly v aminokyselinových profilech plazmy pacientů a zdravých lidí. Vhodným nástrojem pro analýzu biologických vzorků je kapilární elektroforéza s fluorescenční detekcí. Proto byly vyvinuty nové derivatizační a separační podmínky pro stanovení všech proteinogenních aminokyselin v lidské plazmě. Optimalizace a validace metody je předmětem tohoto příspěvku.
