Řešení poskytuje sadu pro stanovení analytu testem využívajícím rekombinantní kvasinky. Sada obsahuje první nádobku s lyofilizovaným živným médiem, druhou nádobku s lyofilizovanými rekombinantními kvasinkami obsahujícími receptor pro analyt a reportérový gen pod transkripční kontrolou tohoto receptoru, a třetí nádobku se zmraženými činidly pro detekci exprese reportérového genu. Rovněž je popsán způsob její přípravy. Dále je poskytnut způsob stanovení analytu ve vodném vzorku s použitím této sady, který obsahuje následující kroky: - vodný vzorek se převede do první nádobky s lyofilizovaným živným médiem pro kvasinky, - vodný vzorek s rozpuštěnými složkami živného média pro kvasinky se převede z první nádobky do druhé nádobky a inkubuje, - činidlo pro detekci exprese reportérového genu se převede ze třetí nádobky do druhé nádobky nebo se inkubovaný vzorek převede z druhé nádobky do třetí nádobky, a detekuje se exprese reportérového genu.

The invention provides a kit for determination of analyte by a recombinant yeast assay, characterized in that said kit contains a first container with lyophilized nutrient medium, a second container with lyophilized recombinant yeast containing a receptor for the analyte and a reporter gene under transcriptional control of this receptor, and a third container with a frozen reagent for the detection of the expression of the reporter gene; as well as a method of preparation thereof. Further, the invention provides a method for determining the analyte in an aqueous sample using the kit according to any one of claims 1 to 3, charakterized in that it comprises the following steps: - the aqueous sample is transferred to the first container with the lyophilized nutrient medium for the yeast; - the aqueous sample with the dissolved components of the yeast nutrient medium is transferred from the first container to the second container with the lyophilized yeast and incubated; - the reagent for the detection of expression of the reporter gene is transferred from the third container to the second container or the incubated sample is transferred from the second container into the third container, and the expression of the reporter gene is detected.
