IR laser ablates nitrocellulose and deposited intact nanoparticles are desorbed. Coupled with ICP MS, nanoparticles can be counted with the single particle detection limit. Enzyme-linked immunosorbent assay (ELISA) with labeling based on the principle of antibody-antigen immunocomplex formation is nowadays a common analytical method. The signal corresponding to immunocomplex formation is usually amplified by an enzyme. In this work, a model assay of the HSA protein (Human Serum Albumin) will be conducted, the enzyme will be replaced by an yttrium upconversion nanoparticle (PLISA, Particle-linked immunosorbent assay). Here we test the possibility of the single particle label detection, furthermore the reaction will be transfered to a nitrocellulose surface and compared to the alternative ULISA (upnconversion particle-linked immunosorbent assay ) detection.
