Abstract
The content of main flavonoids in the root of Scutellaria baicalensis cultivated in Central Europe was evaluated by using new simple RP-HPLC method. The main components of the roots were baicalin-BG (8,12% of dry root mass) and wogonin glucuronide-WG (2,52 %). Five main flavonoids were evaluated for their scavenging ability with DPPH radical and due to limited solubility only two flavonoids were investigated for their ability to scavenge hydroxyl radical by the method aromatic hydroxylation. The total extract was also tested in both the experimental arrangements. In experiments with DPPH only BG and baicalein displayed significant scavenging effect, while the production of OH radical was considerable decreased in the presence of BG and WG. After comparison with results obtained for total extract, it was concluded, that scavenging activity of the extract against DPPH is mainly derived from BG. On the other hand, BG, WG and probably other flavonoids participate in scavenging OH radical.

Obsah hlavních flavonoidů v kořenu Scutellaria baicalensis pěstované v Evropě byl vyhodnocen pomocí nové a jednoduché RP-HPLC metody s gradientem acetonitrilu v mobilní fázi. Hlavními komponentami kořene byl bajkalin-BG (8,12% suchého kořene) a wogonin glukuronid-WG (2,52%). U pěti hlavních flavoidů byla testována jejich antiradikálová aktivita s DPPH a pouze u dvou flavonoidů (vlivem jejich omezené rozpustnosti) byla měřena jejich schopnost vychytávat hydroxylový radikál metodou aromatické hydroxylace. Pro srovnání byl testován i celkový extrakt. V experimentech s DPPH pouze BG a bajkalein vykázal signifikatní zhášecí efekt , zatímco produkce OH radikálů generovaných pomocí UV fotolýzy H2O2 byla snížena v přítomnosti BG a WG. Po porovnání s výsledky získanými s celkovým extraktem, je možno shrnout, že za zhášecí aktivitu extraktu hlavně odpovídá BG. Na druhé straně, BG, WG a pravděpodobně i ostatní flavonoidy působí při zhášení OH radikálu.
