In our laboratory, the method of monophasic action potentials recording using voltage sensitive dye (VSD) di-4-ANEPPS is employed to study changes in electrical activity of mammalian myocardium under various physiological and/or pathological conditions, such as ischemia-reperfusion, preconditioning, etc. Still little is known about the direct effect of voltage sensitive dye of this type on the heart muscle. In the present study, two models of the isolated heart of guinea pig and rabbit perfused according to Langendorf - are compared with respect to changes triggered by VSD application. The hearts are perfused at the constant pressure, spontaneously beating and preparation as well as experiments are carried at 37C. The myocardium of both species undergoes the same procedure before the start of the experiment itself: organ isolation, loading with the dye and washout from the dye. Touch-less 3D electrogram and coronary flow are followed during the whole preparation and experiment. In the course of loading and washout from the voltage-sensitive dye, prominent electrophysiological changes occur (mainly decrease of spontaneous heart rate and changes in the shape of T wave). This electrical impairment is accompanied with decrease in mean coronary flow. However, these changes differ in the two species. In guinea pig, the electrical changes are more prominent and in most cases the RR prolongation does not disappear even after washout period. Also the changes in coronary flow are more accented in guinea pig hearts and are less reversible. It may be concluded that rabbit heart is more resistant to the changes which are triggered with VSD application. Although these changes are partially reversible in guinea pig heart, this model seems to be more sensitive and thus less reliable, especially when the conductive system in the heart is studied.

V naší laboratoři studujeme změny elektrické aktivity savčího myokardu za různých fyziologických i patofyziologických podmínek (ischemie-reperfůze, preconditioning, atd.) registrací monofázických akčních napětí za pomocí napěťově-citlivého barviva di-4-ANEPPSS. O přímých účincích tohoto barviva na srdeční sval se doposud mnoho neví. V naší práci jsme srovnávali účinek barviva na dva modely - izolovaná srdce morčete a králíka perfundovaná dle Langendorfa. Srdce jsou perfundována na konstantním tlaku, spontánně tepou a preparace stejně jako experimenty probíhají při 37C. Obojí srdce procházejí izolací, barvením a výplachem barviva. Celou dobu sledujeme bezdotykově 3D elektrogram a koronární průtok. Významné změny elektrogramu se objevily v době barvení a výplachu barviva - pokles spontánní srdeční frekvence a změny ve tvaru T vlny. Je to doprovázeno poklesem koronárního průtoku. U obou druhů se však změny liší - u morčete jsou více vyznačené a většinou prodloužení RR nezmizí ani po washoutu. Také změny koronárního průtoku jsou markantnější a méně reverzibilní. Lze shrnout, že králičí srdce je odolnější ke změnám spuštěným napěťově-citlivým barvivem. Ačkoliv u morčete jsou tyto změny částečně reverzibilní, zdá se tento model citlivější a tedy méně spolehlivý, obzvláště pro studium převodního systému srdečního.
