Abstract
The prevalence of Borrelia burgdorferi sensu lato in Ixodes ricinus ticks from South Moravia was investigated. A total of 305 ticks (21 larvae, 243 nymphs, 19 females and 22 males) were collected in suburban woods of Pisárky Park (city of Brno) from July to October 2002. All specimens were examined by dark-field microscopy (DFM) for the presence of spirochetes. The mean infection rate was 14.8%. 13.6% of males and 26.3% of females tested were positive for the presence of B. burgdorferi s.l.. Also, of the 243 nymphs, 14.4% were positive and out of 21 larvae 9.5% were positive. Borreliae were identified by polymerase chain reaction- restriction fragment length polymorphism (PCR-RFLP) methods. From a total of 45 DFM positive samples 37 were positive for the presence of borrelian DNA. Isolation attempts resulted in 21 spirochaetal strains. Identification by PCR-RFLP detection revealed genospecies: Borrelia afzelii, B. garinii, B. burgdorferi sensu stricto and B. valaisiana. Four isolated strains showed a mixed population of these genospecies.
Abstract (in Czech)
Na jižní Moravě byla zjišťována promořenost klíšťat druhu Ixodes ricinus bakterií Borrelia burgdorferi sensu lato. Od července do října 2002 bylo v lesích na parkové lokalitě Pisárky v Brně shromážděno 305 klíšťat (21 larev, 243 nymf, 19 samic a 22 samců). Všechny vzorky byly vyšetřeny zástinovou mikroskopií (DFM) na přítomnost spirochet. Průměrná pozitivita byla 14,8%. Bylo pozitivních 13,6% samců a 26,3% samic, 14,4% a 9,5% nymf. Borrelie byly identifikovány metodami PCR-RFLP. Ze 45 DFM pozitivních vzorků bylo 37 pozitivních na přítomnost borreliové DNA. Podařilo se získat 21 izolátů. Výsledkem analýzy PCR-RFLP bylo určení těchto genospecies: Borrelia afzelii, B. garinii, B. burgdorferi sensu stricto a B. valaisiana. 4 izolované kmeny ukázalay směsnou infekci těchto genospecies.
