The bases of nucleic acids contain several quaternary carbon atoms whose detection through the remote protons is difficult or impossible, but that are suitable for the direct carbon detection. Moreover, the network of coupled carbon spins in 13C-enriched compounds causes splitting of the signals in the directly detected dimension. This is a disadvantage in general spectroscopy but it can be used as an easy way of measuring carbon-carbon couplings.
