Expression profiles of peripheral blood CD34+ cells from a healthy donor and from a patient with NHL were compared by cDNA microarray technique during ex vivo granulocyte differentiation. It was found that standard differentiation expression profile of donor’s cells was different from the profile of NHL patient’s cells in terms of gene expression kinetics, counts of genes involved in granulopoiesis, and gene expression levels. The exact cause of found gene expression changes remains to be established. Ex vivo granulocyte differentiation of CD34+ cells is a well established method. Together with cDNA technology it may become a tool for evaluating physiological abnormalities within CD34+ cell population.

Pomocí technologie cDNA microarrays byly srovnány expresní profily CD34+ buněk z periferní krve od zdravého dárce a pacienta s NHL a to během jejich ex vivo granulocytární diferenciace. Mezi sledovanými populacemi buněk byly nalezeny rozdíly v počtu a zastoupení genů zapojených do diferenciačních procesů a v kinetice exprese některých ze sledovaných genů během diferenciace. Otázkou zůstává, co je příčinou této rozdílné genové exprese. Navozená ex vivo granulocytární diferenciace CD34+ buněk je v klinické praxi již zavedenou metodou. Spolu s technologií cDNA microarrays a detailním profilováním genové exprese by se mohla stát nástrojem pro zhodnocení fyziologických abnormalit uvnitř populace CD34+ buněk.
