Abstract
Recent advances in understanding beta-catenin-independent WNT (non-canonical) signaling suggest an increasing complexity, opening the question of how individual non-canonical pathways are induced and regulated. Here we examine whether intracellular signaling components such as beta-arrestin and casein kinase 1 and 2 (CK1 and 2) can contribute to determine signaling specificity in beta-catenin-independent WNT signaling to the small GTPase RAC-1. Our results suggest that the balance between beta-arrestin and CK1/2 determines whether WNT/RAC-1 or other non-canonical WNT pathways are activated.
Abstract (in Czech)
Současné poznatky o Wnt drahách nezávislých na beta-kateninu (tzv. nekanonické Wnt dráhy) ukazují, že jde o komplexní směsici signalizací, což otvírá otázku, jak jsou jednotlivé dráhy indukovány a regulovány. V předložené práci jsme testovali jakým způsobem ovlivňuje beta-arrestin a kasein kinázy 1 a 2 (CK1/2) nekanonickou Wnt dráhu směrem k malé GTPáze RAC-1. Naše výsledky ukazují, že rovnováha mezi beta-arrestinem a CK1/2 určuje, jestli se bude aktivovat RAC-1 nebo jiná nekonanická Wnt dráha.
