Abstract
The aim of this work is in vitro creation of mutant lectins from oportunistic human pathogen Chromobacterium vioalceum. Substitution of amino acids in the lectin structure should lead to changes of lectin sugar specificity. Mutation were focused on the "specificity binding loop" in the position 22-23-24 and in other position 97. The work is going to be followed by structural and functional analysis.
Abstract (in Czech)
Cílem této práce je připravit mutantní lektiny z patogenního organismu Chromobacterium violaceum. Záměna amino kyselin ve struktuře lektinu může vést ke změnám vazebných vlastností lektinu. Mutace byly zaměřeny do místa vazebné specifity v pozici 22-23-24 a do místa v pozici 97. Vazebné vlasnosti mutantních lektinů budou následně charakterizovány strukturně funkčními studiemi.
