Course objectives
Main objective of the course is to introduce students to principals of work with bacterial cells and bacteriophages and presenting them with a set of chosen tasks to equip them with skills needed for solution of various problems they can face in bacterial physiology. Using set of chosen techniques used in physiological laboratories they will familiarize themselves with growth characteristics of bacterial cells and phages, influence of temperature and UV on lysogenic cells and phages, they will prepare crude enzymatic preparation and test its enzymatic activity.
Syllabus
  • Determination of the bacterial proteins and dry-weight. Bacterial growth curve in static culture (using plate counts). One-step growth curve of the polyvalent and virulent staphylopage. Determination of dehydrogenase activity. Preparation of the crude enzymatic preparation (HEP) with phosphatase activity. Determination of phosphatase activity of native cells and HEP. Oxygen consumption (Oxi-Top). Biodegradation. Influence of heavy metals on bacterial growth (Bioscreen).
