Course objectives
The objective of the laboratory course are studies of basic properties of compounds important for biochemistry, and methodological approaches in the study and analytical application of enzymes. Content of the first part includes qualitative and quantitative reactions of carbohydrates, lipids, amino acids, proteins and nucleic acids. The second part is oriented to enzyme kinetics and further studies on properties of free enzymes as well as enzymes which are immobilized in cells.
Syllabus
  • Qualitative reactions of carbohydrates. Carbohydrate thin-layer chromatography.
  • Isolation and identification of lipids from the egg yolk.
  • Separation of lipophilic pigments using adsorption chromatography.
  • Chemical reactions of amino acids and proteins. Paper chromatography of amino acids.
  • Isolation of nucleoprotein from spleen.
  • Gel permeation chromatography in separation of biomolecules.
  • Protein determination.
  • pH effect on enzyme reaction.
  • Determination of the rate of enzyme reaction.
  • Enzyme kinetics.
  • Enzymatic determination of metabolites.
  • The use of ferricyanide as an artificial electron acceptor in the electron transport chain.
  • Closing test.

Assessment methods (in Czech)
První cvičení je věnováno poučení z bezpečnosti práce v biochemické laboratoři. Před každým dalším cvičením se písemným testem ověřuje příprava studenta na danou úlohu. Závěrečný test ověřuje znalosti z principů všech úloh.
