Course objectives
At the end of this course, students should be able to work aseptically, to inoculate and cultivate bacteria, to prepare stained and native microscopic slides, to determine number of bacteria in different samples, to determine indicators of microbial water quality, to use some basic identification methods.
Syllabus
  • 1. Introduction, safety in microbiological laboratory 2. Media for cultivation and their preparation 3. Isolation, inoculation and cultivation of microorganisms 4. Microscopic examination of bacteria (slides preparation, Gram - staining, microscopy and image acquisition) 5. Determination of number of viable bacteria by plate count 6. Bacteria in water - basic microbiological water analysis 7. Determination of the concentration of antibiotics by microbiological methods. Determination of susceptibility of bacteria to antibiotics 8. Identification of bacteria - the use of biochemical tests and standardized Microsystems 9. Determination of the number of phage particles in lysate of bacterial cells 10. Isolation and microscopic examination of bacteria from soil (cellulolytic bacteria, Azotobacter sp., Clostridium sp.), 11. Microorganisms in soil. Negative - staining. Measurments of size of bacterial cell. Microscopic examination of endospores. Phase and Nomarsky contrast. 12. Yeasts, test of vitality, inoculation of microscopic fungi and streptomycetes. 13. Microscopy of microscopic fungi and streptomycetes. 14. Reports, test and microscopic examination/determination of unknown bacterial strain.
