Prerequisites
The course is destined for students of microbiology and other students who are interested in new methods of analysis of microorganisms. The prerequisite is the passed exam of the course Molecular Biology (Bi4020).

Course objectives
The aim of the course is to acquaint students with the principles of selected methods of molecular biology, biochemistry, and biophysics for analysis of microorganisms, with the procedure for selection of a suitable method for solving a real problem, differences in procedures used for bacteria and yeasts and interpreting the results of analyzes. Students will receive specific information about the following processes: 1)Isolation of nucleic acids (NA) from microorganisms and analysis of NA using chromatographic and electrophoresis methods 2)Application of enzymes in microbial genes analysis 3)Sequence analysis of microbial genes, genomes, and proteins 4)Application of hybridisation and polymerase chain reaction in microbiology 5)Methods of study of microbial proteins and metabolites 6)Application of recombinant NA for the study of microorganisms
Learning outcomes
At the end of this course, students should be able to: understand and explain the principles of basic molecular biology, biochemistry and biophysics methods used in microbiology, propose the procedure for solving a real problem, realize differences between methods used for bacteria and yeasts; interpret the results.
Syllabus
  • Methods of microorganisms analysis. Isolation of nucleic acid.
  • Isolation of DNA and mtDNA from yeasts.
  • Fragmentation of DNA molecules. Restriction enzymes.
  • Analysis of nucleic acids using electromigration methods.
  • Analysis of nucleic acids using chromatographic methods.
  • Sequence analysis. Spectrophotometric analysis of nucleic acids.
  • Hybridisation.
  • Polymerase chain reaction (PCR).
  • Methods of protein analysis. Chromatographic determination of microbial metabolites and bacterial cell components.
  • Principles of quantitative analysis.
  • Immunomagnetic and affinity based separation techniques in microorganisms diagnostics.
  • Methods for preparation of recombinant DNA and RNA molecules and their use for the study of microorganisms.

Teaching methods
The teaching methods make use of Powerpoint presentations. The course is followed up by a practical course.
Assessment methods
The course is closed with a test. The test contains 30 questions, which must be answered briefly. Each student must correctly answer 15 questions. The overall result of the exam is evaluated by the examiner. Test if followed by a brief discussion with the examiner (oral part of the exam).
