Course objectives
Light Microscope Construction and Function (Bright Field, Phase Contract Microscopy, Interference-DIC). Replicas for the Study of Surface. Rapid Cytological Methods: Cell Cycle, Nuclear Staining. Embryology: Microsporogenesis, Pollen Grain Development and Structure, Megasporogenesis, Embryo Sac,Fertilisation, Zygote, Embryo and Endosperm Development, Whole Mounts of Cleared Preparations. Histological Methods: Fixation, Dehydration, Infiltration and Embedding, Sectioning, Dyes and Staining. Cryosections and Histochemistry. Fluorescence Microscopy. Documentation.

  • 1. Development of Light Microscope Construction and Function (Bright Field, Köhler Illumination). 2. Replicas for the Study of Surface, Epidermis, Stoma Cells, Subsidiary Cells, Types of Stoma Structure. 3. Rapid Cytological Methods: Meristems, Cell Cycle, Cytoskeleton Organization during Cell Cycle, Nuclear Staining (Lacto-propionic Orcein, Cajal-Brožek Method), Pre-treatment. 4. Androeceum: Stamen, Development and Structure of Anther, Microsporogenesis, Pollen Grain Development and Structure, Pollen Grain Germination, Pollen Grain Viability. 5. Gynaecium: Whole Mounts of Cleared Ovules and Seeds, Megasporogenesis, Embryo and Endosperm Development. Seed Structure. 6. Phase Contract Microscopy, Nomarski DIC. 7. Histological Methods: Fixation, Dehydration, Infiltration and Embedding, Sectioning, Dyes and Staining. 8. Cryosections and Enzyme Histochemistry. 9. Fluorescence Microscopy, Autofluorescence, Fluorochromes, Immunofluorescence. 10. Documentation: Photomicrography, Video, and Distance Measurements.
