Objective: Periodontitis is a complex, multifactorial, chronic inflammatory disease that involves degradation of periodontal structures, including alveolar bone. Vitamin D acts through binding with vitamin D receptor (VDR) and is responsible for regulating bone metabolism and mineralization; it also suppresses the immune system. The aim of this study was to analyze a single nucleotide polymorphism (SNP) in the VDR gene, which may affect the functional activity of the VDR protein, in Czech patients with aggressive (AgP) and chronic periodontitis (CP). Methods: In this case-control study 448 unrelated subjects were included. Genomic DNA of 67 patients with AgP, 305 patients with CP and 76 healthy/non-periodontitis controls were genotyped. For TaqI VDR (rs731236) gene polymorphism the method based on polymerase chain reaction (PCR) followed by restriction digest was used. Results: There was no evidence for a significant association between TaqI VDR polymorphism and CP, while allele frequencies of TaqI VDR polymorphism was significantly different between patients with AgP and non-periodontitis controls (P = 0.015). And although no difference was found in genotype frequencies in patients with AgP, statistically significant results were obtained after comparing the genotypes TT vs CT + CC (P = 0.034), in the subgroup of women (N = 40, P = 0.017). Conclusions: We can conclude relationship between the occurrence of the mutated allele (C) of TaqI VDR polymorphism and the development of aggressive forms of periodontitis, especially in women, but a correlation between mutation and chronic form of periodontitis was not found.
