Sulfur is an essential chemical element for all organisms. Microorganisms can utilize reduced and oxidized sulphur. The most preferred form is sulfate because it is the easiest one to utilize. In this contribution, the most notable pathways are described: assimilatory sulfate reduction, dissimilatory sulfate and sulphur reduction. These pathways often yield toxic hydrogen sulphide, which has a negative impact on human health and the environment. Because of its corrosive properties, H2S also limits some industrial processes. There is a number of microorganisms capable of utilizing H2S, for example, green (Chlorobiacea) and purple (Chromatiaceae) sulfur bacteria. They oxidize hydrogen sulfide, resulting in the creation of elemental sulphur. As the first step in my research, various locations in Slovakia and the Czech Republic had been sampled. However, the subsequent isolation and cultivation of sulphur bacteria from samples proved to be difficult. Different media and cultivation conditions were used with mixed results. To increase the chance of acquiring the positive sample as well as creating a reservoir for their acquisition during winter, four different Winogradsky columns were made.
