Current trends in the rise of multidrug-resistant bacteria put pressure on the standardization of alternative methods for treatments of such complicated infections. One of the promising possibilities is phage therapy and its new direction, which is based on a combination of phages with antibiotics. This strategy builds on the synergy effect between these compounds (phage-antibiotic synergy; PAS) [1]. We focused on staphylococcal phage S10 with podoviral morphology and antibiotics affecting cell wall development (Amoxicillin) and protein synthesis (Gentamicin). Their combined effect was assessed by plaque assay and spectrophotometry in 96-well plate format or higher volumes in personal bioreactors. Obtained results were evaluated by recently published methods, i.e., Virulence Index, and Phage Score. In both environments tested (solid and liquid) were observed a synergy effect of amoxicillin and phage S10. The overall phage virulence in 24 hrs intervals increased with the concentration below the minimum inhibitory concentration. The application of gentamicin resulted in a decrement in phage titer, and the ability of phage propagation was lost at higher concentrations. This study brings new insights into the interaction of phages with antibiotics, which is crucial for their combined application as a treatment in the future.
