Paradiplozoon is the most diverse genus of the Diplozoidae (monogeneans). These parasitesare host specific to cyprinoid fishes. In recent years several new species of Paradiplozoon species from Africa and Asia were described, however, the diversity of Paradiplozoon appears to be underexplored. Therefore, in our research, we aimed to study Paradiplozoon diversity, phylogeny and intraspecific variability of Paradiplozoon with respect to the enormous diversity and endemism of cyprinoids. From 2014 to 2022, in 15 countries in Euro-Mediterranean, Middle East, and Africa, 74 cyprinoid species were sampled and examined for diplozoids. All the specimens were identified morphologically and by molecular data. Phylogenetic analyses and intraspecific genetic variability analyses were performed using the ITS2 region. A total of 10 Paradiplozoon species were collected in the Euro-Mediterranean, Middle East, and Africa, out of which three were described. Paradiplozoon homoion had the widest host range (44 host species, 15 genera) as well as the widest geographical distribution. All investigated Paradiplozoon species were divided into three divergent lineages on the phylogram, suggesting multiple origins of diplozoids in the area. In line with their geographic distribution some generalists of Paradiplozoon species exhibited genetic and morphological intraspecific variabilities. Reconstructed phylogeny supported Paradiplozoon paraphyly, highlighting importance of multilocus molecular studies in revealing diplozoids enigmatic phylogeny. Biogeographical processes had impact on intraspecific variability of Paradiplozoon species. Re-evaluating diplozoid species could expose their hidden diversity, clarify recent lineages diversification, and fill the gaps in their evolutionary history. Czech Science Foundation, project no.
