Analysis of glycosylation is still challenging, not only because of the extreme complexity of glycan structures and their conjugation diversity, but also because of instrumental aspects such as the limits of analytical sensitivity.1 Therefore, glycan analysis by chromatographic or electrophoretic separation methods is very often combined with fluorescence detection in addition to MS.2 The majority of fluorescent labeling employed before separation is based on 2-aminobenzamide, which has several disadvantages such as low labeling yield, poor fluorescence properties, and MS ionization efficiency. Therefore, even after several decades of development of new labels, there is still a need for new labeling tags with improved characteristics.3,4 In this work, we present the synthesis of new cationic 2-phenylpyridine-based tags and their application for oligosaccharide and glycan derivatization via hydrazone formation chemistry. The labeling conditions were optimized using maltooligosaccharide standards, and the labeling efficiency was compared with the labeling using Girard′s reagent P followed by HPLC/MS analysis. Finally, the developed labeling method was applied for CE-MS analysis of N-linked glycans.
