Tick-borne encephalitis virus (TBEV) is a neurotropic orthoflavivirus that invades the central nervous system, causing severe neurological disease and posing a significant public health threat. Traditionally, detecting TBEV in infected cells or animal models relied on labor-intensive secondary approaches, preventing real-time observation of infection and limiting studies of viral spread and host responses. We developed a stable recombinant TBEV expressing TurboGFP, allowing direct visualization of infection in cells, tissue slices, and mice. The reporter virus enables tracking of viral replication, spread, and tropism, including whole brain imaging in 3D using light-sheet microscopy. This tool could also be used for antiviral testing and provides a versatile platform for studying TBEV biology and pathogenesis.
