Anotace
Büyük bal mumu güvesi Galleria mellonella L. larvalarýný laboratuvar ţartlarýnda yetiţtirmek için kullanýlan yapay besine farklý konsantrasyonlarda ilave edilen borik asitin böceđin son evre larvalarý ve puplarýnýn hemolenfindeki protein profili, lipid peroksidasyon düzeyi ve antioksidan savunma sistemine etkisi incelendi. Sodyum dodesilsülfat-poliakrilamid gradient jel elektroforez tekniđi (SDS-PAGGE) ile son evre larvalarýnda yaklaţýk 15 protein bandý saptanýrken puplarda 13 farklý protein bandý gözlenmiţtir. Lipoforin (245 kDa) ve depo proteinleri (80 kDa) gibi bilinen protein bantlarýna ilave olarak 18-260 kDa arasýnda farklý moleküler ađýrlýđýna sahip protein bantlarýna da rastlanmýţtýr. Borik asitin düţük konsantrasyonlarýnda larvalarýn protein profilinde bir farklýlýk gözlenmemiţ olup denenen en yüksek konsantrasyonunda diđer protein fraksiyonlarýna göre daha büyük ve 43 kDa molekül ađýrlýđýnda bir protein fraksiyonu ortaya çýkmýţtýr. Borik asitin yüksek konsantrasyonlarýnda son evre larvalarýnýn hemolenfindeki lipid peroksidasyonu son ürünü olan malondialdehid (MDA) miktarý önemli derecede artmýţtýr. En yüksek borik asit konsantrasyonu (2500 ppm) kontrol larvalarýnýn hemolenfindeki 0.32 ą 0.003 olan katalaz (CAT) aktivitesini 0.04 ą 0.001 mmol/mg protein/dk gibi oldukca düţük bir deđere azaltmýţ, glutatyon S-transferaz (GST) aktivitesini ise 3.30 ą 0.32 den 2.59 ą 0.34 nmol/mg protein/dkya düţürmüţtür. Borik asitin 1250 ppmi puplarýn hemolenfindeki MDA miktarýný 0.30 ą 0.04 den 0.12 ą 0.02 nmol/mg proteine düţürmüţ olup GST aktivitesini etkilememiţtir. Denenen borik asit konsantrasyonlarýnýn tümü puplarýn hemolenfindeki CAT aktivitesini önemli derecede artýrmýţtýr. Bu sonuçlar borik asitin sebep olduđu oksidatif stresin antioksidan savunma sistemini bozan önemli bir faktör olabileceđini göstermekle beraber, antioksidan savunma sisteminin öncelikli enzimleri CAT ve GST aktivitelerindeki önemli azalýţýn meydana getireceđi oksidatif hasar nedeniyle larvalarýn hemolenfinde oksidatif strese karţý koruyucu olarak yeni stres proteinlerinin oluţabileceđini söyleyebiliriz.

Hemolymph protein profiles, lipid peroxidation level and antioxidant defense system of the greater waxmoth, Galleria mellonella (L.) final instars and pupae were investigated by rearing the newly hatched larvae on an artificial diet containing 156, 620, 1250, and 2500 ppm of boric acid. Sodium dodecyl sulphate polyacrylamide gradient gel electrophoresis (SDS-PAGGE) was used to determine the hemolymph protein profiles. Using this technique it was possible to distinguish approximately 15 protein fractions in the final instar larva, and in pupae about 13 protein fractions were observed. In addition to well-defined protein fractions such as lipophorins (245 kDa) and storage proteins (80 kDa), some undetermined protein fractions varying from 18260 kDa were also detected. Although the changes in protein profiles of larvae in lower concentrations of boric acid were initially not detected, later, a clear influence of the highest concentration of boric acid was observed. One protein fraction of molecular weight 43 kDa was significantly bigger after the treatment. No such difference was observed in pupal hemolymph. Lipid peroxidation product, malondialdehyde (MDA) level was significantly increased after high concentrations of boric acid treatment in hemolymph of final instars. Highest concentration of boric acid (2500 ppm) markedly decreased catalase (CAT) activity from 0.32 ą 0.003 in the control to 0.04 ą 0.001 mmol/mg protein/min and glutathione S-transferase (GST) activity from 3.30 ą 0.32 to 2.59 ą 0.34 nmol/mg protein/min in larval hemolymph. Boric acid at 1250 ppm significantly decreased MDA level from 0.30 ą 0.04 in control to 0.12 ą 0.02 nmol/mg protein while GST activity was not significantly affected in pupal hemolymph. However all concentrations of boric acid significantly increased CAT activity in hemolymph of pupae This study suggests that boric acid-induced oxidative stress may be causative factor in deterioration of larval antioxidative defense system and that decreased CAT and GST activities for highest concentration of boric acid may result in appearance of a new stress protein in hemolymph of final instars.
