Abstract
v-Myb oncoprotein of AMV interferes with differentiation of myeloid cells. Differentiation of v-Myb-transformed monoblasts BM2 can be resumed by phorbol ester TPA or histone deacetylase inhibitor trichostatin A. The aim of this work was to explore the effects of the p53 tumor suppressor on proliferation, viability, cell cycle progression, apoptosis and differentiation of BM2 cells. We found that ectopic p53 expression completely suppresses proliferation of BM2 cells by imposing block in the G2 phase of the cell cycle. In addition, p53 induces differentiation of BM2 monoblasts to monocytes, as confirmed by up-regulation of production of reactive oxygen species, expression of CD11b, non-specific esterase and myeloperoxidase activities, and by changes of the intracellular pH. In contrast, frequency of apoptosis was not affected by overexpressed p53 in BM2 cells.
Abstract (in Czech)
Onkoprotein v-Myb blokuje diferenciaci myeloidních buněk. Schopnost diferenciace monoblastů transformovaných v-Myb může být obnovena forbolovým esterem TPA nebo inhibitorem histonových deacetyláz - trichostatinem A. Cílem této práce bylo prozkoumat účinky nádorového supresoru p53 na proliferaci, viabilitu, buněčný cyklus, apoptózu a diferenciaci buněk BM2. Zjistili jsme, že ektopická exprese p53 zcela blokuje proliferaci buněk BM2 z důvodu zastavení buněčného cyklu ve fázi G2. Kromě toho p53 indukuje diferenciaci monoblastů BM2 do stádia monocytů, což jsme potvrdili zvýšenou produkcí reaktivních kyslíkových radikálů, expresí CD11b, aktivitou nespecifických esteráz a myeloperoxidáz a změnami nitrobuněčného pH. Naopak přítomnost exogenního p53 neovlivnila frekvenci apoptózy.
