V originále
Asymmetric dimethylarginine (ADMA) is a posttranslationally modified form of arginine that is generated in all types of cells during the process of degradation methylated proteins. ADMA is a competitive inhibitor of nitric oxide synthase. We report here development of a new HPLC method for the determination of ADMA using coulometric detection. After solid-phase extraction procedure on a strong cation exchange column and evaporation of the eluate under gentle stream of nitrogen at 70 to 75 C the reconstituted samples were derivatised with ortho-phthaldialdehyde reagent containing 3-mercaptopropionic acid. Formed derivatives were analysed on a reversed phase column Hydro-RP using isocratic elution and coulometric detection.
