Abstract
In our study we wanted to examine Ixodes ricinus ticks and different species of mosquitoes for the presence of pathogennic borreliae. We used two method: 1. Dark Field Microscopy (DFM) - detection of spirochetes 2. Polymerase Chain Reaction (PCR) - detection of Borrelia burgdorferi sensu lato DNA We found significantly higher positivity of ticks for borreliae compared to mosquitoes. There was the big discrepance in DFM and PCR positivity. It could be caused by PCR sensitivity (101 copies of DNA) and probably by the presence of other spirochetes. We isolated two bacteria strains from Culex (Culex) pipiens s.l., the first one was identified as B. afzelii, the second one identified as non B.b.s.l.
Abstract (in Czech)
V naší studii jsme chtěli vyšetřit klíšťata druhu Ixodes ricinus a různé druhy komárů na přítomnost patogenních borrelií. Použili jsme dvě metody: 1. Zástinovou mikroskopii (DFM) pro detekci spirochet 2. Polymerázovou řetězovou reakci (PCR) pro detekci DNA Borrelia burgdorferi sensu lato Zjistili jsme statisticky významnou pozitivitu klíšťat na borrelie v porovnání s komáry. Byl velký rozdíl mezi výsledky metod DFM a PCR. To by mohlo být způsobeno citlivostí metody PCR (101 kopií DNA) a pravděpodobně také přítomností jiných spirochet. Izolovali jsme dva bakteriální kmeny z komára Culex (Culex) pipiens s.l., první identifikovaný jako B. afzelii, druhý jako nepatřící do komplexu B.b.s.l.
