QBA are the natural products interesting for their antiproliferative and colour properties. Among them, sanguinarine and chelerythrine are the best recognized. We studied antiproliferative, proapoptotic and colour properties of minor occuring QBA, sanguirubine (SR), chelirubine (CHR), sanguilutine (SL), chelilutine (CHL) and macarpine (MA) in comparison with sanguinarine (SA) and chelerythrine (CHE). All tested alkaloids shoved strong antiproliferative effect on cells of human cancer cell lines that was accompanied with apoptosis induction. Furthermore we observed interesting colour and DNA intercalating properties of MA, SR and CHR. We proved that these alkaloids, especially macarpine, could be used as a new supravital fluorescence DNA stains for both fluorescence microscopy and flow cytometry. Fluorescence microscopy of MA stained cells described the nuclear architecture, chromosomes and apoptotic fragments. Moreover MA binds to DNA stoichiometrically and can rapidly represent the cellular DNA content of living cells at a resolution adequate for flow cytometric cell cycle analysis. Our results show QBA as compounds interesting not only for their antiproliferative activities, but also demonstrate their possible applications as supravital DNA probes for fluorescence microscopy and flow cytometry.
