Markers associated with myelomatous plasma cells (PCs) are mostly known, but only limited information about their relation to prognosis is available. As MGUS pass into MM by multistep transformation process, we try to compare phenotype patterns of these diseases to find parameter with a predictive value in the term of disease progression. Flow cytometric analysis of PC phenotype was done in 112 newly diagnosed patients with MM and 31 MGUS subjects. Bone marrow PCs were identified as CD38+CD138+ cells and expression of CD19, CD20, CD27, CD28, CD56 and CD117 was analysed on these PCs in 5-colors setting. Flow cytometry is a sensitive technique which allows detection of a few PCs in MGUS and MM cases, however phenotypic characterization and discrimination of normal and malignant PCs should be more important then their number. Higher number of CD19+ normal PCs correlate with MGUS as higher number of CD56+ myelomatous PCs correlate with MM. CD27 seems to have a predictive value for progression of disease and should be monitored for its possible prognostic information as well. ER -
