The aim of this study was to detect and characterise L. monocytogenes in rippening cheese from retail market in the Czech Republic. A total of 142 samples originating from the Czech and foreign producers were tested in the year 2007. The detection and enumeration of L. monocytogenes were performed according to ČSN EN/ISO 11290 - 1, 2. L. monocytogenes isolates were characterised by serotyping using the slide agglutination method and multiplex PCR assay and typing has been performed by restriction analysis using pulsed-field gel electrophoresis (PFGE). L. monocytogenes was detected in 12 samples of czech origin: in 17% (4/24) of smeared cheese samples, 15% (7/48) of blue-veined cheese and 1% (1/70) of cheese with the mould on the surface. All isolates belonged to serotype 1/2a. Using macro-restriction analysis, 3 different pulsotypes have been detected. Although positive L. monocytogenes samples were detected on retail level, none of them exceeded the count of 100 CFU.g-1. Specific pulsotypes have been detected for each of the individual cheese producers. |
