Proteomic analysis of tetraspanin-deficient extracellular vesicles
Anotace
Tetraspaniny jsou transmembránové proteiny zapojené do různých buněčných procesů a hrají klíčovou roli v extracelulárních vezikulech (EV). EV jsou malé dvojitou membránou ohraničené struktury, které zprostředkovávají mezibuněčnou komunikaci přenosem proteinů, lipidů a genetického materiálu. Zájem vědecké komunity o EV neustále roste. Nedávné studie zkoumaly účinky knockoutu jednoho tetraspaninu z membrány …více
Abstract
Tetraspanins are transmembrane proteins involved in various cellular processes and play a crucial role in extracellular vesicles (EVs). EVs are small membrane-bound structures that mediate intercellular communication through the transfer of proteins, lipids, and genetic material. The scientific community’s interest in EVs is steadily increasing. Recent studies have explored the effects of single tetraspanin …více
Zadání práce
The student will:
1) create a comprehensive literature review to summarize the existing knowledge regarding the role of tetraspanins in EVs;
2) conduct a bioinformatic analysis of mass spectrometry (MS) data from EVs obtained from three different cell lines in which the key tetraspanins (CD9, CD63, and CD81) have been knocked out using KNIME platform and specific R libraries (e.g. DEP, QFeatures). Subsequently, she will follow with the biological interpretation of the results, including gene ontology and pathway analysis.
These findings will form the foundation for designing subsequent experiments aimed at elucidating the individual contributions of tetraspanins to EV biogenesis, cargo selection, cell targeting, and uptake.
