Anotace

Abstract

Subarachnoid hemorrhage (SAH) is a type of stroke. It is a severe medical condition with serious consequences and high mortality. The initial 72 hours after the onset, referred to as “Early brain injury” (EBI), is driven by complex pathophysiological processes. Central role plays an inflammation. This thesis investigates the variability of the inflammatory response based on the severity of bleeding …více

Zadání práce
Subarachnoid hemorrhage (SAH) is a severe medical condition affecting around 8 out of 100 000 individuals per year. High mortality is caused either by the onset of bleeding initially or by subsequent complications such as re-bleeding, early brain injury (EBI), cerebral vasospasm (CVS), and delayed cerebral ischemia (DCI). Inflammation represents pathology which may contribute to all the SAH-related complications. One of the most studied inflammatory markers is Interleukin-6 (IL-6), which is associated with the severity of bleeding as well as with the final outcome. However, similarly to other pro-inflammatory cytokines, there is high interindividual variability in IL-6 levels. The goal of this thesis is to characterize the variability of IL-6 in the experimental model of SAH. The student will use a rat endovascular perforation model and basic laboratory methods. Timeline: 1st semester (2022): literature review, microsurgical technique training 2nd semester (2023): SAH model training, surgeries, sample collection, laboratory analyses. If necessary, modification of the experimental design (model switch) 3rd semester (2023): surgeries, sample collection, laboratory analyses, and evaluation. 4th semester (2024): final evaluation, thesis preparation, and defense
