Prerequisites
Basic chemical nomenclature in Czech (symbols and names of elements, nomenclature of inorganic compounds – acids, bases, salts, oxides), basic types of organic compounds.

Course objectives
Subject is complementary to the Biochemistry lecture and seminars ABCHp and ABCHs. Students become familiar with the background and equipments of chemical laboratory. They will acquire basic laboratory skills, learn to perform simple laboratory operations and become familiar with the processing of biological material, especially urine and blood. During experiments students verify findings discussed in theoretical lessons (lectures and seminars). Part of the practises is focused on the issues of methods used in clinical biochemistry and their practical use.
Learning outcomes
After completing the course, the student will:
- be familiar with safety and health protection at work in the laboratory;
- be able to calculate the concentration of solutions, pipette, prepare or dilute a solution of a given concentration;
- be acquainted with the work with biological material and its processing in the laboratory of clinical biochemistry;
- be able to describe the principles of sampling biological material and its processing
- be familiar with orientation examination of urine using diagnostic strips and with the use of selcted POCT analyzers
- be able to work with basic instruments in the biochemical laboratory, eg. pH meters, spectrophotometers;
- understand the principle and implementation of selected methods used in clinical biochemistry (optical, chromatographic, electrophoretic, immunochemical methods).
Syllabus
  • Introductory information, safety rules. Preparing solutions of the given concentration.
  • The measurement of pH of various solutions. pH-metres.
  • Biological material. Examinations in clinical biochemistry and factors influencing the results. Biochemical analyzers. determination of selected analytes using POCT analyzers.
  • Spectrophotometry, absorption curve. Determination of glucose.
  • Chromatography. Gel chromatography. Adsorption chromatography of plant pigments (silicagel). HPLC simulation.
  • Electrophoretic and immunochemical methods. Proteins in blood serum and urine. Electrophoresis of blood serum proteins. ELISA.

Teaching methods
Homework theoretical preparation, practical exercises, presentation of laboratory methods and instruments.
Assessment methods
Full attendance in all lessons (making up all missing and justified lessons), completion of all lab reports is the condition for the course-unit credit. Obtaining of course-unit credits of practicals is the pre-requisite for registration to the examination.

The course is taught annually.
Teacher's information
Attendance in practicals is obligatory. Laboratory coat, laboratory gloves and overshoes are necessary. Student is obliged to prepare himself for each lesson. Students that are not sufficiently prepared will not be admitted in participating in the lesson. All absences must be justified through the Department of study affairs. After being absent the student must make up for the lesson. Student may have only one unexcused absence during the semester.
