Course objectives
At the end of the course students should be able to: - explain basic terms used in microbiology, clarify main differences among individual groups of microorganisms, mention their basic taxonomic classification and enumerate their typical species;
- describe individual work instruments used in microbiological laboratory and interpret their characteristic functions;
- prepare culture media for isolation, growth and storage of microorganisms;
- apply basic microbiological methods (isolation, cultivation and identification of microorganisms, inoculation, observation of microorganisms by microscope etc.);
- understand an importance of sterile work in microbiological laboratory;
- characterize generally an occurrence of microorganisms in environment (soil, water), their contribution to nutrients flow in environment and their significance for man (etiological agens of infectious diseases, commensals, using of microorganisms in industry and in research, bioremediation)
Syllabus
  • 1. Theory: Labour safety. Protocols. Introduction to microbiology. Microbiology as a branch of science. Historical milestones of microbiology and its significance today. Importance of microorganisms. Basic classification of microorganisms. Fundamental principles of work with microorganisms, characterization of working tools and instrumentation in microbiological laboratory. Significance of sterilization and aseptic work. Practical work: Labour tools sterilization.
  • 2. Theory: Cell. Characterization of prokaryotic cell. ABC of bacterial taxonomy. Types of culture media. Practical work: Preparation of culture media for microorganisms cultivation.
  • 3. Theory: Methods of microorganisms inoculation, cultivation, isolation (streaking) and identification. Practical work: Inoculation, cultivation and isolation (streaking) of microorganisms.
  • 4. Theory: Microscopy and microorganisms I. - Principles of work with microscope. Kinds of microscopes. Microscopic techniques. Mistakes during microscopy. Service of microscope. Practical work: Microorganisms observation using optical microscope - bacterial cells morphology.
  • 5. Theory: Microscopy and microorganisms II. - native, fixed and permanent preparations, methods of microorganisms staining. Assessment of microorganisms’ colonies morphology. Practical work: Microorganisms observation using optical microscope - microorganisms staining (Gram staining, etc.).
  • 6. Theory: Effect of environment on microorganisms. Cell cycle of prokaryotic organisms. Practical work: Methods of cell amount assessment in microbial culture (microscopic cell amount assessment, cell amount assessment using cultivation and nephelometric cell amount assessment).
  • 7. Theory: Importance of bacteria for man I. - pathogenic and non-pathogenic bacteria, using of microorganisms in research. Microorganisms ecology I. - microorganisms and water. Practical work: Tentative determination of bacterial water pollution by means of psychrophilic and mesophilic bacteria cultivation.
  • 8. Theory: Importance of bacteria for man II. - using of bacteria in industry, bioremediation. Microorganisms ecology II. - microorganisms and soil, microorganisms and food. Practical work: Isolation of soil bacteria fixing air nitrogen using culture media. Microbiological analysis of food (yogurt) by cultivation. Microscopical observation of bacterial cultures isolated from food (yogurt).
  • 9. Theory: Biologically active substances - dyes, antibiotics, vitamins, growth factors. Diffusion method. Practical work: Detection of dyes and antibiotics influence on microorganisms growth by bacteria agar plate cultivation.
  • 10. Theory: Characterization of eukaryotic cell. Yeasts: their importance for man (fermentative processes, model organisms, pathogens). Importance of the yeast Saccharomyces cerevisiae. Practical work: Cultivation and microscopy of the yeast cells. Saccharomyces cerevisiae in different cell cycle stages. Vital staining of Saccharomyces cerevisiae cells - vital test.
  • 11. Theory: Introduction to veterinary microbiology. Other microscopic organisms - viruses, fungi, parasites.
  • 12. Practical microbiology - excursion to chosen microbiological workplace (will be specified during semester).
  • 13. Internet sources, microorganisms collections, control of protocols.

Teaching methods
theoretical preparation, laboratory course
Assessment methods
Individual laboratory course during whole semester are obligatory for students (accepted is one unexcused absence). Control of filled work protocols by teacher is necessary condition for successful completion of the course. Students acquire credit after test with 20 questions.
